TripAdvisor's Popularity Index, which measures overall traveller satisfaction, put the lesser-known property in Downtown Burj Khalifa ahead of 370 other hotels across the emirate.
Visitors have described the Al Mazil as a "hidden gem" that has "a fresh and modern décor with the right touch of old Arabic".
Last year, the hotel was also voted the Top Business Hotel in Dubai on the website's Business Popularity Index.
The index is based on customer ratings, reviews and other sources, a statement released on Sunday said.
"While Southern Sun has garnered numerous industry accolades in the past few years, receiving this vote of confidence from our guests is a strong incentive to our staff to keep up the good work and maintain the reputation we have been building since we entered the market in 2007," said Richard Weilers, Managing Director Southern Sun Middle East.
Al Manzil Hotel is one of two 4-star hotels operated by the hospitality group, Southern Sun, which manages properties across Africa.
